MC1R receptors, activated by α-MSH and its analogues, are inhibited by natural antagonists present in the skin, the AGRPs (AGouti-Related Protein). These propteins have a phenylalanine in he key position which plays an essential role in binding to MC1R receptors. They thus inhibit melanogenesis.  

 The lightening efficiency of α-MSH is high than the lightening efficiency of wele-know lightening agents such as hydroquinone, kojic acid, arbutine, and magnesium ascorbyl phosphate (VCPMG).
